If I leave a back link for my Manta directory listing, will it help to move up that listing?

It depends upon the type of link.If it is do-follow it will certainly help you in case of higher SERP results.If it is no-follow although it will no such link juice but can drive some traffic for you.

Yes, it depends on link characteristics such as it should have "do-follow" quality so that Google considered it for ranking. Also, it should be natural looking and on high PR site.

Not only Mantra directory listing, most of the Directory listing are doing do-follow links. so it may helpful to you improve your page rank. Also you get some little traffic. Submitting to DMOZ will be high impact in your ranking. Because it's most trusted Directory right now.
