How to check if my site is over optimized ???Is their any tools through which I can check whether my site is over optimized or not ??

To avoid over-optimization, use variation of the anchor text. That would definitely help. Any anchor text over 20% of the anchors(other than your brand name), may get penalized.

Over-optimization is a process of google that is particularly difficult to figure out by user..because Google calculates rankings by considering thousands of different factors.so we can do only read first google search engine guidelines and optimize site proper way..

Optimization of any website is very important to promote it, all activities like link building strategies and all promotional activities are useful to optimize any website, but when huge spamming and unethical way of marketing process or link building process take place then it come under overly optimization, although optimization is good but still over optimization can lead to harm website.

Over Optimization happens when your website is considered too good by Google - either in terms of a sudden volume of back links, or because of heavy on-page optimization. Google considers that your website optimization is beyond acceptable limits; your website will be red-flagged and automatically restricted or penalized.

There isn't any tool for verifying over optimization. You need to perform your optimization based on various keywords and following webmaster guidelines which will enable safe seo for your website.

I Think you can ask to Google by webmaster tool your work is Over Optimization or Not..?

Check out the keywords density to know whether your site is over optimize or not. Optimize your keywords within the limits to avoid over optimization.
